Objectives:
the elaboration of a innovative treatment protocol in the traumatic medullary lesions, which should include calpaine inhibitors and angiogenesis factors, which promotes the survival of the neurons, oligodendrocytes and angiogenesis, resulting in functional recovery of traumatic medullary lesions;
the accomplishment of applicable experimental models for the study of synergic influences of calpaine inhibitors + vascular trophic factors;

OBJECTIVES
  • elaborating  innovative treatment protocol in the traumatic medullary lesions, which should include calpaine inhibitors and angiogenesis factors, which promotes the survival of the neurons, oligodendrocytes and angiogenesis, resulting in functional recovery of traumatic medullary lesions;
  •  developing applicable experimental models for the study of synergic influences of calpaine inhibitors + vascular trophic factors;
  • elaborating clinical study of this well-defined pathology with performing technical means, on carefully selected cases.
DESCRIPTION            The goals of the project are: the evaluation of the recovery of the neurological function after a traumatic medullary lesion, of patients treated with a combination of calpaine inhibitors (for limiting the apoptosis-like events) and of angiogenesis factors (to favor the development of a capillary bed that supports neuroregeneration). This combined effect will be evaluated within the experimental model (in vivo) through neurological function testing (quantification of motor and sensory deficit), histological analysis and histochemistry of the samples at established intervals.

Novelty and originality of the project are given by:

  • the quantification of the combined protective effects on the in vivo experimental model of the calpaine inhibitors and of the angiogenesis factors administrated post-traumatic;
  • the correlation of the functional status with histological and histochemical aspects;
  • treatment protocol of the traumatic intramedullary lesions, which contains calpaine inhibitors and angiogenesis factors;
  • the evaluation of the synergic effects of the calpaine inhibitors + angiogenesis factors, for the elaboration and optimal dose of the compounds;
  • the identification of the mechanisms involved in secondary lesions associated with traumatic medullary lesions;
  • the induction of in vivo angiogenesis.

The complexity of the project is given by:

  • the accomplishment of experimental models in vivo for the evaluation of the combined effects of calpaine inhibitors and vascular trophic factors;
  • the emphasize of the role of the ` HGF-cMET complex in motility, mitogenesis and cell morphogenesis;
  • the conception of the in vivo angiogenesis induction model;
  • the evaluation of the histopathology in animal models;
  • the accomplishment of the quantifying model of the protective effects combined with the in vivo experimental model of calpaine inhibitors and of angiogenesis factors administrated post-traumatic;
  • the correlation of the functional status with the histological and histochemical aspects;
  • the elaboration of the treatment protocol of the traumatic medullary lesions, which should contain calpaine inhibitors and angiogenesis factors- determining the optimal concentration and the mode of administration.
